Is spatial sound a DbD setting or something done outside the game?
It's a Windows feature. You can enable in the sounds settings of your PC. It should make the sounds of games more "3D"

Does that feature work with any headphones or do the headphones need to specifically be marked as having that capability?
As someone who plays a lot of Spirit, I think it's really overrated. It's usually not an output quality issue keeping you from hearing things in this game. Proximity and occlusion affect sounds a ton in this game. I have audiophile headphones and an amplifier.
Survivors can be screaming at me from 8 meters away and then suddenly go completely silent at 8.5 meters. No amount of spatial sound, gear, etc., will change the proximity at which sounds play. An equalizer program can help with clarity, but I've found spatial sound takes away as much as it gives you. It may make a positive difference with your specific rig though. Never hurts to try both.
